摘要
The structural stability of cell assemblies is a necessary prerequisite for in vitro culturing. Swell and damaged cross-linked net structures are the main factors influencing the stability. A stable cell and gelatin assembly for in vitro culturing was developed based on the chemical reaction and diffusion mechanics of a glutaraldehyde and hydrogel mixture composed of chitosan and gelatin, using a 0.8%~1% glutaraldehyde solution as the cross-linker and controlling the cross-linking time for 15~60 s. Tests show that the formatted 40~50 μm protective layer on the outside of cell assembly built using a low concentration of glutaraldehyde as a cross-linker, is not toxic to cells and can be degraded. The layer maintains the macro stability of the cell assembly during the long term incubation time. The low concentration glutaraldehyde solution is suitable as the process reagent for structure stability.
