N2 - BACKGROUND: Mesenchymal stem cells (MSCs) can be induced into osteoblast, condrocytes, adipocytes, cardiomyocytes and nerve cells in vitro. MSCs characterized by multi-lineage differentiation, easy separation and culture have been widely used in stem cell therapy and tissue engineering research. SIRT1, a protein deacetylase widely distributing in mammals, has been proved to control the proliferation and differentiation of MSCs through regulating related genes. OBJECTIVE: To summarize the effects of SIRT1 on MSCs differentiation, focusing on signal pathways and other related factors regulation. METHODS: The keywords of “SIRT1, mesenchymal stem cells, differentiation” were searched in CNKI, PubMed and Elsevier for related papers. Irrelevant, outdated and repetitive articles were excluded, and the remaining 49 articles were included for further analysis. RESULTS AND CONCLUSION: MSCs have the potential of self-renewal and multi-lineage differentiation via Wnt, transforming growth factor β and other signal pathways. SIRT1 influences the self-renewal and multi-lineage differentiations of MSCs, including up-regulation of osteogenic, chondrogenic and myogenic differentiations and down-regulation of adipogenic differentiation, and the underlying mechanism is to regulate the expression, activity and location of intermediate proteins in signal pathways and differentiation-related transcription factors.
