Abstract
A molecularly imprinted polymer (MIP) with high selectivity was prepared using methacrylic acid (MAA) as monomer, ethylene glycol dimethacrylate (EDMA) as cross-linker and L-pazufloxacin as template. The binding capacity of MIP to the template is higher than that of blank polymer. Scatchard analysis show that the MIP recognized template with two classes of binding sites. The multiple-sites binding model was used to calculate the dissociation constant Kd and apparent maximum number Qmax of binding sites. Kd,1 and Qmax,2of the binding sites with high affinity are 23.08 μmol/L and 3.65 μmol/g, while Kd,2 and Qmax,2 of the binding sites with low affinity are 0.43 mmol/L and 27.28 μmol/g respectively. The selective binding experiment for substrates shows that the affinity and selectivity for pazufloxacin are higher than that for L-norfloxacin and ofloxacin.
